First off all, the new site updates are epic! Here's my thoughts I'd like to share:

Spotlighting

I don't want to be mean or cruel to anyone, but I personally hate Spotlighting already. I cam back after a few hours, and got 47 notifications. ALL of them were spotlights. Personally I think a user should be limited to 5-10 spotlights In 24 hours, so you don't have stuff like that happen so much. Or, they could be smart notifications, like say 5 spotlights happened, it would have one notification.

Jet Stream

It's an okay idea. I do have to go to a separate page though, but I'm a lazy person, so yea. It's good IMO.

New Format

Epic. Enough said.

Bio

I've been waiting for so long. I think it's good to show what interests you have and whatnot.

User Blocks

50/50 for me. It can get very rude and hateful users off your back, but they can change and be nice. Also, I think there should be a way to prevent people from blocking you mindlessly.

Userpage format

I wish that the [More Posts] thing was still here, rather than only highlights. But again, it's not a bad inclusion.

Seeing who you are following

Finally. Now I know every single person who I'm following so I don't always forget. Thank goodness.

New Forums

Not bad at all! Now it's not much of a trash can mess. But I do think the search tags should of stayed.
